The fluorescence response of chlortetracycline-loaded human neutrophils is modulated by prostaglandin El, but not by cyclic nucleotides

چکیده
Human neutrophils preloaded with chlortetracycline, commonly used as a probe of membrane-bound calcium, demonstrate a prompt decrease in fluorescence when exposed to surface stimuli such as the chemotactic peptide fMet-Leu-Phe. The fluorescence response was highly sensitive to preincubation with prostaglandin Ex. This effect was apparently not due to elevated levels of CAMP sink exogenous dibutyrylCAMP did not alter the chlo&tracycline fluorescence response to fMet-Leu-Phe. This is one of the few instances of prostaglandin 131 affecting neutrophils at physiologic concentrations, dissociated from changes in cellular cyclic nucleotide levels.

متن کاملCYCLIC NUCLEOTIDES CONTROL DIFFERENTIATION OF HUMAN MONOCYTES INTO EITHER HIGHLY ACCESSORY CELLS OR MACROPHAGES
Human peripheral blood monocytes have been found to undergo a transitory state of high accessory activity before they fully become macrophages. Time kinetics were done to follow this accessory potential. Studying the regulation of accessory activity, we have found that monocyte derived accessory cells (m-AC) pass through two phases of development, both of which are adversely controlled by ...

Ca 2+, Mg2+-ionophores X537A and A23,187 (10-r-10 -6 M) induced the release of adenine nucleotides adenosine diphosphate (ADP, adenosine triphosphate (ATP), serotonin, /3-glucuronidase, Ca 2+, and Mg 2+ from washed human platelets. Enzymes present in the cytoplasm or mitochondria, and Zn 2+ were not released. The rate of ATP and Ca 2+ release measured by firefly lantern extract and murexide dye...
متن کاملThe effects of epinephrine and prostaglandin E-1 on cyclic adenosine 3':5'-monophosphate levels in WI-38 fibroblasts.
1. Intact WI-38 human diploid lung fibroblasts were shown to respond to epinephrine (3 X lop7 M) and prostaglandin El (PGE1) (0.57 X lop7 M) with increased cyclic adenosine 3’: 5’-monophosphate (cyclic AMP) levels. Corticotropin (1.25 units per ml), insulin (0.02 to 200 milliunits per ml), and glucagon (5 X lo-’ M) were ineffective. Although the response to PGE, was not clearly maximal at conce...

In contrast, 15(S)-15-methyl-prostaglandin E1 and prostaglandin I2 did not alter the rate or the total amount of superoxide production by human neutrophils stimulated with either phorbol myristate acetate or arachidonic ac...
